Can I do this on a smart trainer?

So let's say I had a smart trainer (eg. Kickr) and a Garmin Edge 520. Could I go to a site like Ride with GPS, map out a route up Alpe d'Huez, download the GPX file of that route and upload it to the Edge 520 as a course, and then use the Edge 520 to control the resistance on the Kickr to simulate a climb of Alpe d'Huez, all from the comfort of my Canadian home?

The 520 should have FE-C protocol/communication and allow route workouts and simulation to terrain.

Yes, but as others said the videos are pretty key. If you are interested in General rides Tacx trainer suite is clunky, but it allows street view from google maps and an overlay map to be used. If you are interested in famous climbs -- veloreality has a great selection.
